I know location is always a large factor but I'm asking...
What else does it take to make a successful store? What kind of funds should I have available when I start out? Which markets are used most by people? What items are the best to sell/buy? How should the store appear?
After having spent the $20,000 on the 4 weeks minimum for the store, I'd have $40,000+ available to spend on anything to get it up and running. Of course, I wouldn't mind going out and finding things myself.

The store need to be nice. There are some things that attract me to a store.
Open layout - things aren't hidden from my first glance at the store
Blocks behind trade signs - I can see the exact block I need instead of wasting time reading all the actual sign
Good prices - at or under average market price. Overpriced goods=turn away
Stock- The store needs to have plenty of what I want. Make sure to have backup stock for the store.
I've always used redmarket first and then went on to blue but people have different opinions.-

To add on to what @Jacob43365 has said, cheaper prices and good deals are what people are looking for. For example, you sell 32 baked potatoes for 20$.

Keep things organized and prioritized.
The front of your store, the first thing people see, should be the most competitive deals. Netherstars, exp, diamonds, emeralds, etc.
Keep items and building materials separate. Don't sell potions and cobblestone in the same place, it's cluttered and annoying.
Take advantage of your space and build your shop as large as possible and keep it as open and spacious as you can

Good deals on nether stars, EXP, diamonds, and emeralds were my main drivers of profit when I owned Station10. Albeit in a weird location (yellow2, second right), I had 3 other users nearby who advertised their stores when I wasn't online. I then tried to keep my prices competitive to drive interest when I wasn't online, and it turned a pretty penny.
Organization is key, and noting what you are buying and what's being sold is important too.
But seriously, you have a great location at the first row of green2. With competitive pricing and solid advertising, you should be able to make lots.

I used to run a spawnshop and the most important thing to do is keeping a large quantity of each stock. Knowing that your shop is sure to have what they are looking for in stock, the customers are a lot more likely to come back. This takes a lot of work but it does pay off xD

I have just started renting a spawn shop(Station54) Blue Market. I would say the best thing to do is to make sure that you have stock of everything you have decided to sell, because if you don't loyal customers will be forced to look elsewhere. Pricing must be good and advertise every opportunity you get. Try and create some excitement. Having TP is also great so that you can instantly transport customers to your shop so they wont have to walk past other shops on the way.

Shop layout, ease of use and dependability are more important than prices, in my experience. You can charge more if you have a great layout, its easy to use and the stuff is always in stock.
I also strongly believe in having no walls on the front of the store. Wide open entrance.-
